Field Validation of Toxicity Tests to Evaluate the Potential for Beneficial Use of Produced Water

This study investigated potential biological effects of produced water contamination derived from occasional surface overflow and possible subsurface intrusion at an oil production site along the shore of Skiatook Lake, Oklahoma. We monitored basic chemistry and acute toxicity to a suite of standard aquatic test species (fathead minnow-Pimephales promelas, Daphnia pulex, Daphnia magna, and Ceriodaphnia dubia) in produced water and in samples taken from shallow groundwater wells on the site. Toxicity identification evaluations and ion toxicity modeling were used to identify toxic constituents in the samples. Lake sediment at the oil production site and at a reference site were also analyzed for brine intrusion chemically and by testing sediment toxicity using the benthic invertebrates, Chironomus dilutus, and Hyallela azteca. Sediment quality was also assessed with in situ survival and growth studies with H. azteca and the Asian clam, Corbicula fluminea, and by benthic macroinvertebrate community sampling. The produced water was acutely toxic to the aquatic test organisms at concentrations ranging from 1% to 10% of the whole produced water sample. Toxicity identification evaluation and ion toxicity modeling indicated major ion salts and hydrocarbons were the primary mixture toxicants. Sections of this report include: symposium activity, staff activity, document analysis program, text-retrieval program, institute activity, etc. It is believed that as large, complete collections of documents become available in digital libraries, users will demand complete interaction with the information; document access mechanisms will have to grow beyond keywords and full-text searches to include browsing, searching of images, and searching on basis of abstract concepts. It is proposed to study the microform document conversion process, including image preprocessing, recognition, postprocessing for extracting information, and natural language techniques. Characterization of algorithms will allow generation of a system that automatically adapts to a wide range of image quality, thereby allowing large-scale conversion efforts. It is proposed to focus first on the NSF Antarctic database (approx. 55,000 documents).

Proposal for a High Energy Nuclear Database

We propose to develop a high-energy heavy-ion experimental database and make it accessible to the scientific community through an on-line interface. This database will be searchable and cross-indexed with relevant publications, including published detector descriptions. Since this database will be a community resource, it requires the high-energy nuclear physics community's financial and manpower support. This database should eventually contain all published data from Bevalac and AGS to RHIC to CERN-LHC energies, proton-proton to nucleus-nucleus collisions as well as other relevant systems, and all measured observables. Such a database would have tremendous scientific payoff as it makes systematic studies easier and allows simpler benchmarking of theoretical models to a broad range of old and new experiments. Furthermore, there is a growing need for compilations of high-energy nuclear data for applications including stockpile stewardship, technology development for inertial confinement fusion and target and source development for upcoming facilities such as the Next Linear Collider. To enhance the utility of this database, we propose periodically performing evaluations of the data and summarizing the results in topical reviews.

Compliance status report for the Waste Isolation Pilot Plant

The US Department of Energy (DOE) is responsible for the disposition of transuranic (TRU) waste generated through national defense-related activities. Approximately 53,700 m{sup 2} of these wastes have been generated and are currently stored at government defense installations across the country. The Waste Isolation Pilot Plant (WIPP), located in southeastern New Mexico, has been sited and constructed to meet the criteria established by the scientific and regulatory community for the safe, long-term disposal of TRU and TRU-mixed wastes. This Compliance Status Report (CSR) provides an assessment of the progress of the WIPP Program toward compliance with long-term disposal regulations, set forth in Title 40 CFR 191 (EPA, 1993a), Subparts B and C, and Title 40 CFR {section}268.6 (EPA, 1993b), in order to focus on-going and future experimental and engineering activities. The CSR attempts to identify issues associated with the performance of the WIPP as a long-term repository and to focus on the resolution of these issues. This report will serve as a tool to focus project resources on the areas necessary to ensure complete, accurate, and timely submittal of the compliance application. This document is not intended to constitute a statement of compliance or a demonstration of compliance.

Geohydrologic data have been collected in the Los Medanos area at the US Department of Energy`s proposed Waste Isolation Pilot Plant (WIPP) site in southeastern New Mexico since 1975 as part of a study evaluating the feasibility of storing defense-associated nuclear wastes within the bedded salt of the Salado Formation of Permian age. Drilling and hydrologic testing have identified three principal water-bearing zones above the Salado Formation and one below that could potentially transport wastes to the biosphere if the proposed facility were breached. The zones above the Salado are the contact between the Rustler and Salado Formations and the Culebra and Magenta Dolomite Members of the Rustler Formation of Permian age. The zone below the Salado Formation consists of channel sandstones in the Bell Canyon Formation of the Permian Delaware Mountain Group. Determinations of hydraulic gradients, directions of flow, and hydraulic properties were hindered because of the negligible permeability of the water-bearing zones. Special techniques in drilling, well completion, and hydraulic testing have been developed to determine the hydrologic characteristics of these water-producing zones.

The Impact of Glucuronidation on the Bioactivation and DNA Adduction of the Cooked-Food Carcinogen 2-Amino-1-methyl-6-phenylimidazo[4,5-b] pyridine in vivo

UDP-glucuronosyltransferases (UGTs) catalyze the glucuronidation of many different chemicals. Glucuronidation is especially important for detoxifying reactive intermediates from metabolic reactions, which otherwise can be biotransformed into highly reactive cytotoxic or carcinogenic species. Detoxification of certain food-borne carcinogenic heterocyclic amines (HAs) is highly dependent on UGT1A-mediated glucuronidation. 2-Amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP), the most mass abundant carcinogenic HA found in well-done cooked meat, is extensively glucuronidated by UGT1A proteins. In humans, CYP1A2 catalyzed N-hydroxylation and subsequent UGT1A-mediated glucuronidation is a dominant pathway in the metabolism of PhIP. Therefore, changes in glucuronidation rates could significantly alter PhIP metabolism. To determine the importance of UGT1A-mediated glucuronidation in the biotransformation of PhIP, UGT1A proficient Wistar and UGT1A deficient Gunn rats were exposed to a single 100 {micro}g/kg oral dose of [{sup 14}C]-PhIP. Urine was collected over 24 h and the PhIP urinary metabolite profiles were compared between the two strains. After the 24 h exposure, livers and colon were removed and analyzed for DNA adduct formation by accelerator mass spectrometry. Wistar rats produced several PhIP and N-hydroxy-PhIP glucuronides that accounted for {approx}25% of the total amount of recovered urinary metabolites. The Waste Isolation Pilot Plant (WIPP) is a research and development facility for the demonstration of the permanent isolation of transuranic radioactive wastes in a geologic formation. The facility was constructed in southeastern New Mexico in a manner intended to meet criteria established by the scientific and regulatory community for the safe, long-term disposal of transuranic wastes. The US Department of Energy (DOE) is preparing an application to demonstrate compliance with the requirements outlined in Title 40, Part 191 of the Code of Federal Regulations (CFR) for the permanent disposal of transuranic wastes. As mandated by the Waste Isolation Pilot Plant (WIPP) Land Withdrawal Act of 1992, the US Environmental Protection Agency (EPA) must evaluate this compliance application and provide a determination regarding compliance with the requirements within one year of receiving a complete application. Because the WIPP is a very complex program, the DOE has planned to submit the application as a draft in two parts. This strategy will allow for the DOE and the EPA to begin technical discussions on critical WIPP issues before the one-year compliance determination period begins. This report is the first of these two draft submittals.

Reducing Open Cell Landfill Methane Emissions with a Bioactive Alternative Daily

Methane and carbon dioxide are formed in landfills as wastes degrade. Molecule-for-molecule, methane is about 20 times more potent than carbon dioxide at trapping heat in the earth's atmosphere, and thus, it is the methane emissions from landfills that are scrutinized. For example, if emissions composed of 60% methane and 40% carbon dioxide were changed to a mix that was 40% methane and 60% carbon dioxide, a 30% reduction in the landfill's global warming potential would result. A 10% methane, 90% carbon dioxide ratio will result in a 75% reduction in global warming potential compared to the baseline. Gas collection from a closed landfill can reduce emissions, and it is sometimes combined with a biocover, an engineered system where methane oxidizing bacteria living in a medium such as compost, convert landfill methane to carbon dioxide and water. Although methane oxidizing bacteria merely convert one greenhouse gas (methane) to another (carbon dioxide), this conversion can offer significant reductions in the overall greenhouse gas contribution, or global warming potential, associated with the landfill. What has not been addressed to date is the fact that methane can also escape from a landfill when the active cell is being filled with waste. Federal regulations …

In May 2002, the United States Department of Energy (DOE) signed Assistance Instrument Number DE-FC09-02CH11109 with the Medical University of South Carolina (MUSC) to support the Environmental Biosciences Program (EBP). This funding instrument replaces DOE Assistance Instrument Number DE-FC02-98CH10902. EBP is an integrated, multidisciplinary scientific research program, employing a range of research initiatives to identify, study and resolve environmental health risks. These initiatives are consistent with the MUSC role as a comprehensive state-supported health sciences institution and with the nation's need for new and better approaches to the solution of a complex and expansive array of environment-related health problems. The intrinsic capabilities of a comprehensive health sciences institution enable MUSC to be a national resource for the scientific investigation of environmental health issues. EBPs success as a nationally prominent research program is due, in part, to its ability to task-organize scientific expertise from multiple disciplines in addressing these complex problems Current research projects have focused EBP talent and resources on providing the scientific basis for risk-based standards, risk-based decision making and the accelerated clean-up of widespread environmental hazards. WESF hot cells waste minimization criteria hot cells window seals evaluation

WESF will decouple from B Plant in the near future. WESF is attempting to minimize the contaminated solid waste in their hot cells and utilize B Plant to receive the waste before decoupling. WESF wishes to determine the minimum amount of contaminated waste that must be removed in order to allow minimum maintenance of the hot cells when they are placed in ''laid-up'' configuration. The remaining waste should not cause unacceptable window seal deterioration for the remaining life of the hot cells. This report investigates and analyzes the seal conditions and hot cell history and concludes that WESF should remove existing point sources, replace cerium window seals in F-Cell and refurbish all leaded windows (except for A-Cell). Work should be accomplished as soon as possible and at least within the next three years.

This report describes the conceptual design of a system the Department of Energy (DOE) may implement for compliance with the requirement to control access to the disposal site. In addition, this report addresses the scheduling process for control of inspection, maintenance, and periodic reporting related to Long Term Monitoring which addresses the monitoring of disposal system performance, environmental monitoring in accordance with the Consultation and Cooperation Agreement between the DOE and the state of New Mexico, and evaluation of testing activities related to the Permanent Marker System design. In addition to access control addressed by this report, the controlling or cleaning up of releases from the site is addressed in the Conceptual Decontamination and Decommissioning Plan. The monitoring of parameters related to disposal system performance is addressed in the Long Term Monitoring Design Concept Description. Together, these three documents address the full range of active institutional controls planned after disposal of the TRU waste in the WIPP repository.

Southwest Regional Partnership on Carbon Sequestration

The Southwest Partnership on Carbon Sequestration completed its Phase I program in December 2005. The main objective of the Southwest Partnership Phase I project was to evaluate and demonstrate the means for achieving an 18% reduction in carbon intensity by 2012. Many other goals were accomplished on the way to this objective, including (1) analysis of CO{sub 2} storage options in the region, including characterization of storage capacities and transportation options, (2) analysis and summary of CO{sub 2} sources, (3) analysis and summary of CO{sub 2} separation and capture technologies employed in the region, (4) evaluation and ranking of the most appropriate sequestration technologies for capture and storage of CO{sub 2} in the Southwest Region, (5) dissemination of existing regulatory/permitting requirements, and (6) assessing and initiating public knowledge and acceptance of possible sequestration approaches. Results of the Southwest Partnership's Phase I evaluation suggested that the most convenient and practical ''first opportunities'' for sequestration would lie along existing CO{sub 2} pipelines in the region. Action plans for six Phase II validation tests in the region were developed, with a portfolio that includes four geologic pilot tests distributed among Utah, New Mexico, and Texas. Campbell)--The NIF amplifiers require 3380 Nd-doped laser glass slabs; continuous glass melting methods will be used for the first time to manufacture these slabs. The properties of the laser glasses are summarized and the novel continuous melting method is described. (2) Diffractive Optics for the NIF (J. A. Britten)--We have fabricated demonstration diffractive optics according to the NIF baseline design at full scale, via wet-chemical etching of patterns into fused silica. We have examined the effects of dip-coated sol-gel antireflection coatings on the performance of these optics, and have concluded that diffractive optics should remain uncoated to minimize laser-induced damage to downstream optics and to maximize environmental stability. We have also demonstrated the feasibility of combining all diffractive structures required by NIF, which vary over orders of magnitude in lateral and vertical scales, onto a single surface. (3) Producing KDP and DKDP Crystals for the NIF Laser (A. K. Burnham)--Rapid-growth KDP has overcome most of the hurdles for production of boules for NIF switch crystals and doublers, but some improvements in process reliability at the tripler's 3{omega} damage threshold are needed. …
