Dna Profiling of Captive Roseate Spoonbill (Ajaia Ajaja) Populations As a Mechanism of Determining Lineage in Colonial Nesting Birds.

Roseate spoonbills are colonial nesting birds with breeding grounds extending from the United States Gulf coast to the pampas of Argentina. The U.S. population suffered a severe bottleneck from 1890 to 1920. The population's recovery was slow and partially credited to migrations from Mexican rookeries, but a gene pool reduction would be expected. Five polymorphic Spoonbill autosomal short tandem repeat (STR) loci [three (GAT)n, one (AAAG)n and one (GT)n] and one Z/W-linked microsatellite exhibiting sex-specific dimorphism were isolated and characterized. The Z/W-linked STR locus accurately confirmed the sex of each bird. Allelic profiles for 51 spoonbills obtained from Dallas (Texas), Fort Worth (Texas) and Sedgwick County (Kansas) zoos revealed a non-continuous distribution of allele frequencies, consistent with the effects of a population bottleneck. Allelic frequencies also differed significantly between the isolated zoo populations. Although extra-pair copulations were suspected and difficult to document, zoos commonly used observational studies of mating pairs to determine familial relationships among adults and offspring. STR parentage analysis of recorded family relationships excluded one or both parents in 10/25 cases studied and it was further possible to identify alternative likely parents in each case. Principal objectives of my research were to: (1) report for the first time that coelomocytes are able to reduce NBT dye and confirm the presence of lysozyme-like activity in earthworm; (2) develop a standard methodology for determination of NBT reduction and lysozyme-like activity in earthworms; (3) compare NBT reduction and lysozyme-like activity in earthworms with those of murine and human cells and fluids; and (4) demonstrate the sensitivity of earthworm NBT reduction and lysozyme-like activity as the assays using matrics in refuse-derived fuel fly ash (RDFF) and CuSO4.

Blood Pressure Regulation During Simulated Orthostatism Prior to and Following Endurance Exercise Training

Cardiovascular responses and tolerance to an orthostatic stress were examined in eight men before and after eight months of endurance exercise training. Following training, maximal oxygen consumption and blood volume were increased, and resting heart rate reduced. Orthostatic tolerance was reduced following training in all eight subjects. It was concluded that prolonged endurance training decreased orthostatic tolerance and this decrease in tolerance appeared associated with attenuated baroreflex sensitivity and alterations in autonomic balance secondary to an increased parasympathetic tone noted with training.

Autophosphorylation and Autoactivation of an S6/H4 Kinase Isolated From Human Placenta

A number of protein kinases have been shown to undergo autophosphorylation, but few have demonstrated a coordinate increase or decrease in enzymatic activity as a result. Described here is a novel S6 kinase isolated from human placenta which autoactivates through autophosphorylation in vitro. This S6/H4 kinase, purified in an inactive state, was shown to be a protein of Mr of 60,000 as estimated by SDS-PAGE and could catalyze the phosphorylation of the synthetic peptide S6-21, the histone H4, and myelin basic protein. Mild digestion of the inactive S6/H4 kinase with trypsin was necessary, but not sufficient, to activate the kinase fully

In Vitro Studies of Nuclear Changes in Mammalian CNS Neurons Subjected to Rapid Acceleration Impact Injury

An in vitro model of Rapid Acceleration Impact (RAI) Injury was used to study the effects of multiple impact (220 g/impact, 3-5 sec intervals) trauma on cultures of murine CNS cells. Investigations with spinal cord cultures showed that 1) multiple impacts delivered tangential to the plane of cell growth caused neuronal death (12% after 3 impacts to 46% after 10 impacts); 2) multiple impacts delivered normal to the plane of cell growth were much less effective (8% dead after 10 impacts); 3) most neuronal death occurred within 15 minutes after injury 4) morphological changes observed included increased nuclear prominence and somal swelling; and 5) pretreatment with ketamine (0.1mM) reduced cell death from 51 to 14% and reduced somal swelling. Identical studies performed on cortical cultures revealed minimal differences between the two tissues in their response to multiple tangential impacts.

Optimization of In Vitro Mammalian Blastocyst Development: Assessment of Culture Conditions, Ovarian Stimulation and Experimental Micro-Manipulation

Factors currently at the forefront of human in vitro fertilization (IVF) that collectively influence treatment success in the form of blastocysts development were investigated during early mammalian embryology with concentration on infertile patients presenting with diminished ovarian reserve or preliminary ovarian failure. A novel experimental technique, Graft Transplant-Embryonic Stem Cells (GT-ESC) was introduced in the mouse model, as the first inclusive approach for embryo selection in IVF treatments resulting in successful graft integration of sibling cells, stage-dependent (day 4) blastocysts. E-Cadherin-catenin bonds play an integral role in trophectoderm cell viability and calcium removal, inducing disruption of cell-to-cell bonds at the blastocyst stage was detrimental to continued blastocyst development. One of the leading methods for embryo selection for uterine transfer in human IVF is application of pre-implantation genetic screening (PGS) methods such as next generation sequencing (NGS). Female patients <35 y do not benefit from this treatment when outcome is measured by presence of fetal heart beats at 10 weeks of gestation. Patients 35-37 y benefit from PGS with no significant difference of outcome based on form of PGS method utilized. The Passage of Sodium-24 and Rubidium-86 Across the Blood-Brain Barrier System of Canines at Low Body Temperatures

To evaluate the blood-brain barrier system in the pathogenesis of an irreversible hypothermic state in dogs, concentrations of 2 4 Na and 86Rb were measured at body temperatures ranging from 37 0 C to 160 C. A suppression of transport of sodium was demonstrated, followed by an increase as the temperature was lowered. The concentration of rubidium ion increased in concentration as the temperature fell. These data indicate there may be a temperature threshold below which the blood-brain barrier system fails to maintain the internal environment of the central nervous system. The intimate relationship of several brain stem nuclei with the cerebro-spinal fluid indicates they may be at risk during profound cooling.

Sensory neocortex contains a significant number of inhibitory neurons that use gamma-aminobutyric acid (GABA) as their neurotransmitter. Functional roles for these neurons have been identified in physiological studies. For example, in primary somatosensory cortex (SI), blockade of GABAa receptors with bicuculline leads to expansion of receptive fields (RFs). The magnitude of RF enlargement varies between SIpopulations of GABAergic neurons were identified by labeling specific calcium binding proteins.

Informing Conservation Management Using Genetic Approaches: Greater Sage-Grouse and Galápagos Short-Eared Owls as Case Studies

Small isolated populations are of particular conservation interest due to their increased extinction risk. This dissertation investigates two small wild bird populations using genetic approaches to inform their conservation. Specifically, one case study investigated a Greater Sage-grouse (Centrocercus urophasianus) population located in northwest Wyoming near Jackson Hole and Grand Teton National Park. Microsatellite data showed that the Jackson sage-grouse population possessed significantly reduced levels of neutral genetic diversity and was isolated from other Wyoming populations. Analysis with single nucleotide polymorphisms (SNPs) and microsatellite data provided further evidence that the population's timing of isolation was relatively recent and most likely due to recent anthropogenic habitat changes. Conservation recommendations include maintaining or increasing the population's current size and reestablishing gene flow with the nearest large population. The second case study investigated the genetic distinctiveness of the Floreana island population of the Galápagos Short-eared Owl (Asio flammeus galapagoensis). Mitochondrial DNA sequence data did not detect differences across nine island populations, yet microsatellite and morphometric data indicated that limited gene flow existed with the population and surrounding island populations, which appeared asymmetric in direction from Floreana to Santa Cruz with no indication of gene flow into Floreana. Monitoring Watershed Health in the Upper Trinity River Basin, North Central Texas

This study conducts watershed analysis using biological and geo-spatial techniques. Incorporating landscape features with biological attributes has been shown to be an effective method of monitoring environmental quality within watersheds. In situ biomonitoring using the Asiatic Clam, Corbicula fluminea, habitat suitability, and water quality data were evaluated for their potential to describe ecological conditions in agricultural and urban areas within the Upper Trinity River watershed. These data were analyzed with GIS to identify effects of land use on ecological conditions. C. fluminea downstream of point source effluents was effective detecting in-stream toxicity. Ambient toxicity appears to have improved in the Trinity, although urban influences limit aspects of aquatic life. No association between habitat quality and land use was identified.

Some Responses of Bacillus subtilis Spores to Glutaraldehyde

Bacillus subtilis (ATCC 19659) were damaged by exposure to various concentrations of glutaraldehyde, as shown by decreased germination rates. The damage caused was repaired or otherwise obviated by the presence of sodium lactate in the holding medium. When two different salts of lactic acid were compared for ability to overcome the effect of glutaraldehyde, it was found that calcium salt of lactate was more effective than the sodium salt. The damage repair system involved l-alanine, lactate and either the sodium or calcium ions. The study involved in determining the difference in efficiency of spore repair was due to an organic or an amino acii snowed that the presence of two carboxylic functional groups did not effectively alter the reactivity.

Greater, Lesser, Guessers: A Look into the Hybridization of Greater and Lesser Prairie-Chickens

My thesis focuses on the conservation consequences of the hybridization of Lesser Prairie-Chickens in Kansas. Specifically, examining how past land management practices altering the species ranges impact the distinctiveness of Lesser Prairie-Chickens. Each chapter is an individual publication that addresses if the Greater and Lesser Prairie-Chicken are distinct when applying the morphological and biological species concepts. Chapter 2 compares the evolutionary history and morphological construct of Lesser Prairie-Chickens and other Galliformes using morphometric analysis. Chapter 3 uses low-resolution microsatellite data to reflect recent changes at the population level. This study aims to observe the Greater and Lesser Prairie-Chicken using the morphological and biological species concepts, two of the many species concepts, to determine the distinctiveness and rate of hybridization for these closely related species.

Toxicity of Chromium and Fluoranthene From Aqueous and Sediment Sources to Selected Freshwater Fish

Research efforts in aquatic toxicology have historically centered on the chemical analyses and toxic effects of waters to aquatic organisms. More recently, sediment-source toxicity has been explored, with efforts concentrated on establishing sensitive and accurate methodologies. This study focused on the toxicity of trivalent chromium, hexavalent chromium, and fluoranthene to Pimephales promelas, Ictalurus punctatus, and Lepomis macrochirus. Test fish were exposed to both water-borne and sediment-source toxicants for 96 hours (h) and 30 days (d). A 96-h and 30-d LC50 (mg/L Cr, ug/L Fluoranthene) was determined for each fish species exposed to aqueous toxicants. In addition, 96-h and 30-d LC50s were determined for each fish species from sediment chromium concentrations (mg/kg) and sediment fluoranthene concentrations (ug/kg). Although lethality endpoints were used throughout this research, acute effects other than mortality were determined for Lepomis macrochirus exposed to hexavalent chromium. Lethal toxicity values (96-h and 30-d LC50 and their 95% confidence limits) for trivalent chromium could not be determined since trivalent chromium concentations above 6.0 mg/L could not be obtained at water pHs compatible with these fish species. Trivalent chromium addition to test waters at pHs compatible with fish survival resulted in a chromium precipitate that was not lethal to test fish. …

Organisms synthesize stress proteins in response to a variety of stressors. The 68/70-kDa proteins (synonymous to the 72/73-kDa proteins) have shown to be the most promising stress proteins, and have been proposed as a biomarker of general organismal stress. The 68/70-kDa proteins were used in an antigen/antibody based approach to determine the duration of the stress protein response of Pimephales promelas following an acute exposure to copper sulphate.

Responses of Selected Texas Fishes to Abiotic Factors, and an Evaluation of the Mechanisms Controlling Thermal Tolerance of the Sheepshead Minnow

Low oxygen tolerances of ten fishes were estimated using an original nitrogen cascade design, and reciprocally transformed to express responses as ventilated volume necessary to satisfy minimal oxygen demand (L·mg O2^-1). Values ranged from 0.52 to 5.64 L·mg^-1 and were partitioned into three statistically distinct groups. Eight stream fishes showed moderately high tolerances reflecting metabolic adaptations associated with stream intermittency. Juvenile longear sunfish and two mollies comprised the second group. High tolerance of hypoxia may allow juvenile sunfish to avoid predation, and mollies to survive harsh environmental oxygen regimens. The sheepshead minnow was the most tolerant species of low oxygen, of those examined, explaining its presence in severely hypoxic environments.

Ecosystem Services and Sustainability: A Framework for Improving Decision-Making in Urban Areas

Ecosystem services are the varied goods and benefits provided by ecosystems that make human life possible. This concept has fostered scientific explorations of the services that nature provides to people with the goal of sustaining those services for future generations. As the world becomes increasingly urban, ecosystems are reshaped, and services are degraded. Provisioning and regulating ecosystem services, landscape planning, decision making, and agricultural systems and technologies play a distinctive role in feeding and sustaining the expanding urban population. Hence, the integrated assessment of these coupled components is necessary to understand food security and sustainable development. Nevertheless, frameworks that incorporate ecosystem services, urbanization, and human wellbeing are still scarce due to several conceptual and methodological gaps that challenge this assessment. As a consequence, these frameworks are not operationalized, and ecosystem services rarely receive proper attention in decision making. This dissertation seeks to improve our understanding of the role of ecosystem services at the landscape level and provides an approach for operationalizing decisions that affect sustainable practices and human wellbeing.

The Development of Potential Therapeutic Anti-Myosin S2 Peptides that Modulate Contraction and Append to the Heart Homing Adduct Tannic Acid without Noticeable Effect on Their Functions

This dissertation aimed to explore the S2 region with an attempt to modulate its elasticity in order to tune the contraction output. Two peptides, the stabilizer and destabilizer, showed high potential in modifying the S2 region at the cellular level, thus they were prepared for animal model testing. In this research, (i) S2 elasticity was studied, and the stabilizer and destabilizer peptides were built to tune contraction output through modulating S2 flexibility; (ii) the peptides were attached to heart homing adducts and the bond between them was confirmed; and (iii) it was shown that minor changes were imposed on the modulating peptides' functionality upon attaching to the heart homing adducts. S2 flexibility was confirmed through comparing it to other parts of myosin using simulated force spectroscopy. Modulatory peptides were built and computationally tested for their efficacy through interaction energy measurement, simulated force spectroscopy and molecular dynamics; these were attached to heart homing adducts for heart delivery. Interaction energy tests determined that tannic acid (TA) served well for this purpose. The stoichiometry of the bond between the TA and the modulating peptides was confirmed using mass spectroscopy. Conservation, Connectivity, and Coexistence: Understanding Corridor Efficacy in Fragmented Landscapes

Conservation corridors, areas of land connecting patches of natural land cover, are frequently cited and implemented as a restorative strategy to counteract fragmentation. Current corridor ecology focuses on experimental corridor systems or designed and built conservation corridors to assess functionality. Such systems and designs are typically short, straight swaths of homogenous land cover with unambiguous transitions between patches. Quantifying the degree to which amorphous landscape configurations, tortuosity, and heterogeneity of land cover and land uses within the corridor has on functional connectedness is a crucial yet overlooked component of corridor efficacy studies. Corridor literature lacks a robust and repeatable methodology for delineating existing landscape elements, recognizing arbitrary edges, and identifying the start and end of ambiguous transitions between the patches and corridor. Using a set of landscapes being studied as part of a global assessment of corridor efficacy, I designed a workflow that standardizes the boundary of corridor-patch interfaces. The proposed method is a quantitative and repeatable approach that minimizes the subjectivity in corridor delineations. This research investigates the degree to which the existence of a corridor modifies the structural and functional connectivity between patches connected by a corridor compared to an intact reference area.

The Microbial Retting Environment of Hibiscus Cannabinus and Its Implications in Broader Applications

Fiber-yielding plants is an area of increased interest due to the potential use in a variety of green-based materials. These biocomposites can be incorporated into multiple uses; for example, to replace building materials and interior vehicular paneling. The research here aims to focus in on the crop Hibiscus cannabinus for utilization into these functions. H. cannabinus is economically attractive due to the entire process being able to be accomplished here in the United States. The plant can be grown in a relatively short growth period (120-180 days), and then processed and incorporated in a biocomposite. The plant fiber must first be broken down into a useable medium. This is accomplished by the retting process, which occurs when microbial constituents breakdown the heteropolysaccharides releasing the fiber. The research aims to bridge the gap between the primitive process of retting and current techniques in molecular and microbiology. Utilizing a classical microbiological approach, which entailed enrichment and isolation of pectinase-producing bacteria for downstream use in augmented microbial retting experiments. The tracking of the bacteria was accomplished by using the 16S rRNA which acts as “barcodes” for bacteria. The Population Genetic Structure of Quadrula aurea (Bivalvia: Unionidae), A Threatened Freshwater Mussel in Central Texas

The anthropogenic alteration of riverine ecosystems has led to declines in the abundance and diversity of freshwater mussels (Bivalvia: Unionoida) worldwide. Central Texas is home to a diverse freshwater mussel fauna including three candidates for federal listing under the Endangered Species Act. Surveys conducted over the last few decades suggest many of the endemic freshwater mussel species in Texas exist in small isolated populations that may be vulnerable to the deleterious effects of genetic diversity loss. Microsatellite primers from two closely related species were used to identify a set of genetic markers that functioned in the Golden Orb (Quadrula aurea). Microsatellite markers were then applied to document the population genetic structure of Q. aurea within and among three connected river drainages in southeastern Texas. Gene flow within existing Q. aurea populations appears high indicating little potential for genetic issues stemming from isolation and inbreeding. Two weakly divergent admixed populations were identified occupying the San Antonio and Guadalupe/San Marcos rivers. Population genetic structure was related to river basin affiliation, but results for environmental factors were unresolved.
